چکیده

OBJECTIVES Cadmium (Cd) is ubiquitous in the environment and exposure through food and water as well as occupational sources can contribute to a well-defined spectrum of disease. The present study was undertaken to evaluate the role of hesperetin (Hp) in alleviating the Cd induced biochemical changes in rats. MATERIALS AND METHODS During the experiment, male Wistar rats were injected with Cd 83 mg/kg day) subcutaneously alone or with oral administration of Hp 840 mg/kg day) for 21 days. RESULTS In Cd treated rats the levels of plasma lipid peroxidation (LPO) markers: thiobarbituric acid reactive substances (TBARS) and lipid hydroperoxides (LOOH) were significantly increased while the levels of plasma non-enzymatic antioxidants: reduced glutathione (GSH), vitamins C and E were significantly decreased in Cd administered rats. Administration of Hp along with Cd significantly decreased the level of LPO markers with elevation of non-enzymatic antioxidants in plasma. In vitro studies on the effect of Hp on scavenging 1,1-diphenyl-2-picrylhydrazyl (DPPH*), 2,2-azinobis-(3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS+), superoxide anion (O*-), hydroxyl (OH*) radicals and reducing power also confirmed the free radical scavenging and antioxidant activity of Hp. In addition to that, ascorbic acid, butylated hydroxyl toluene was used as the reference antioxidant radical scavenger compounds. Thus, the observed effects are due to the free radical scavenging and antioxidant potential of Hp. Interestingly, among the different concentrations, tested 50 microM of Hp showed the highest antioxidant and free radical scavenging activities when compared to other concentrations. CONCLUSIONS The result of these findings provides further evidence to the neutraceutical and pharmaceutical potentials of Hp.

Vulnerability of Prepubertal Mice Testis to Iron Induced Oxidative Dysfunctions In Vivo and Functional Implications

Background The present study describes the susceptibility of prepubertal testis of mice to prooxidant induced oxidative impairments both under in vitro and in vivo exposure conditions. MaterialsAndMethods Following in vitro exposure to iron (5,10 and 25 M), oxidative response measured in terms of lipid peroxidation and hydroperoxide levels in testis of pre pubertal mice (4 wk) was more robust c...

Naringin attenuates diabetic retinopathy by inhibiting inflammation, oxidative stress and NF-κB activation in vivo and in vitro

Objective(s): Naringin, an essential flavonoid, inhibits inflammatory response and oxidative stress in diabetes. However, whether naringin has beneficial effects on diabetic retinopathy (DR) remains unknown. Materials and Methods: Streptozotocin (STZ, 65 mg/kg) was intraperitoneally injected into male rats (8 weeks old weighting 200-250 g) to establish diabetic model, then naringin (20, 40 or 8...

The antioxidant effect of hesperetin and nano-hesperetin on activity of catalase and superoxide dismutase enzymes in the hippocampus of animal model of Parkinson's disease

Background and objectives: Hesperetin flavanone is a natural bioflavonoid found abundantly in citrus fruits with antioxidant and anti-inflammatory properties. Nano sizing techniques improve the bioavailability of poorly soluble drugs such as hesperetin. Main feature of Parkinson's disease is the degeneration of dopaminergic neurons in the substantia nigra. The rate of oxidative...

Therapeutic Effect of Deferasirox and Glycine on Chronic Cadmium Toxicosis in Rats

Objective: It has been shown that deferasirox can reduce blood and tissues lead content in animal models. In this study the effect of deferasirox alone or combined with glycine as an antioxidant was evaluated in chronic cadmium toxicosis in rat. Methods: Male wistar albino rats were exposed to 200 ppm cadmium in the drinking water for 3 weeks and treated thereafter with deferasirox (140 mg/kg),...
